- First inspect each synapse the AI identified in the segment to make sure they were properly labeled and the points are well placed. To edit synapse annotations (placing new annotations/deleting incorrect annotations), hold CTRL + left click both points.

- When placing new points, always annotate the Axon first and the Dendrite second. The red point will always place first and the blue second (Axon=red, Dendrite=blue).

- Start off by looking through all the synapses the AI identified. You can open the 3rd drop down menu at the top of the screen labeled “Synapses” to quickly jump to each annotation or by selecting them individually in 2D or 3D by right clicking on them.

- The quickest way to find missed synapses when labeling an Axon is to look at the 3D mesh and focus on any boutons it has. Boutons may have more than one synapse.

- When annotating a Dendrite, first look through all visible spine heads in the 3D mesh, then look through the 2D EM stack to find any synapses along the trunk of the dendrite the AI might have missed.

- When you’re confident all synapses are correct, first hit “Save edges”, then click “Next Segment”. You can go back to previous segments to make changes, just make sure to save edges afterward.
- If you find anything other than an Axon or Dendrite (glia, blood cell, etc), click on the 3rd drop down menu labeled “Synapses” and click “Delete All Points”, then save
